چکیده

The study was designed to assess the effects and optimal range of osmotic pressure of citrate egg yolk extender (CEYE) for cryopreservation of buffalo bull semen. Semen samples of four Nili-Ravi buffalo bulls, kept for artificial insemination (AI) purposes, were collected at weekly interval for five weeks. Five solutions of tri-sodium citrate dihydrate and D (-) fructose of varying osmotic pressures (255, 265, 275, 285 and 295 mOsm/kg) were used to prepare citrate egg yolk extender. The semen samples were frozen at -196°C using 20 x 10spermatozoa/0.5ml straw. The postthaw evaluation was carried out in terms of spermatozoa motility, viability, acrosomal integrity, plasma membrane integrity (PMI), spermatozoa DNA integrity and lipid peroxidation. The buffalo bull spermatozoa motility was significantly (P<0.05) affected by varying osmotic pressures and the spermatozoa were more motile (50.3 ± 4.2 %) in the semen samples extended with CEYE at an osmotic pressure of 295 mOsm/kg compared to 275, 265 and 255 mOsm/kg. Other spermatozoa characteristics including viability, acrosomal integrity, PMI, DNA integrity and lipid peroxidation were remained same in all the treatments groups. The osmotic pressure of 285-295 mOsm/kg was considered to be most suitable for most of the spermatozoa characteristics for the cryopreservation of buffalo bull semen.

Effect of Non-enzymatic Antioxidants in Extender on Post-thaw Quality of Buffalo (bubalus Bubalis) Bull Spermatozoa

The objective of the present study was to determine the effect of non-enzymatic antioxidants (vitamin C or E) in tris-citric acid buffer (TCA) on post-thaw quality of buffalo (Bubalus bubalis) bull spermatozoa. Split pooled buffalo bull ejaculates were diluted in TCA egg yolk glycerol extender containing either vitamin C (TCAC), vitamin E (TCAE) or without antioxidant (TCAN) at 37°C. Extended s...

The present study was designed to see whether adding glutathione to tris-citric egg yolk extender improves the quality of buffalo bull spermatozoa stored at 5°C for five days. Semen collected from Nili-Ravi buffalo (Bubalus bubalis) bulls (n = 5) was diluted (37C; 10 x 10 motile spermatozoa per ml) with extender having glutathione (0.0, 0.5, 1.0, 2.0 and 3.0mM). The extended semen was cooled fr...

Cryopreservation of in situ cool stored buffalo (Bubalus bubalis) epididymal sperm

The objectives of this study were to investigate the effects of testis-epididymis cool storage on viability and progressive motility of buffalo epididymal sperm (EP) and to compare the influences of two basic semen extenders on post thaw EP viability and progressive motility. Abattoir collected buffalo testicles were allotted to three storage times (0 h: n = 10; 24 h: n = 10 and 48 h: n = 12). ...
